Relationship between DNA Damage and Monoubiquitinated Histone H2A in Coke Oven Workers

  • [Objective] To examine the relationship between DNA damage and monoubiquitinated histone H2A in coke oven workers.

    [Methods] A total of 106 coke oven workers(exposed group)and 73 water treatment workers(control group)were recruited in this study. The levels of urinary 1-hydroxypyrene(1-OHP)were determined using high performance liquid chromatography. Comet assay and Western blot were used to detect the DNA damage and the monoubiquitylation of histone H2A, respectively.

    [Results] The mean Olive tail moments(OTM)(0.46)and urinary 1-OHP concentrations(0.34 μmol/mol Cr)in the exposed group were significantly higher than those in the control group(0.37 and 0.21 μmol/mol Cr, respectively, P<0.05). But the median of expression of monoubiquitinated histone H2A(0.61)in the exposed group were significantly lower than that in the control group(0.91, P<0.05). By taking OTM as the dependent variable, the results of multiple linear regression analysis showed that urinary 1-OHP and the relative expressions of monoubiquitinated histone H2A fit the equation as independent variables, and the coefficient of determination R2 was 0.383.

    [Conclusion] DNA damage and urinary 1-OHP are closely related with the relative expression of monoubiquitinated histone H2A in the coke oven workers.
