归脾丸溶液对苯染毒小鼠学习记忆及海马形态学的影响

Effect of Gui-pi Decoction on Learning Memory and Hippocampal Morphology in Mice Exposed to Benzene

  • 摘要: 目的 观察归脾丸溶液对苯中毒小鼠记忆功能的影响,初步揭示归脾丸溶液改善苯中毒小鼠记忆功能的机理。

    方法 72只昆明小鼠,随机分为空白对照组、单纯染毒组、西药组、归脾丸溶液低(2 mg/kg& #183;d)、中(4 mg/kg& #183;d)、高(8mg/kg& #183;d)剂量组共6组,每组12只;西药组服用利血生。用Morris水迷宫系统进行行为学测定,并进行海马神经细胞形态学检查和外周血检测。

    结论 与染毒组相比,归脾丸溶液高、中、低剂量组水迷宫定位导航寻找平台时间减少,差异均有统计学意义(P<0.01);归脾丸溶液高剂量组的水迷宫空间搜索跨台次数、目标区域游泳时间增加,差异有统计学意义(P<0.01或P<0.05)。海马组织中神经细胞形态及数量有明显差异。与西药组相比,归脾丸溶液高、中剂量组的血红蛋白明显升高,差异均有统计学意义(P<0.01或P<0.05)。

    结论 归脾丸溶液可改善苯中毒小鼠的记忆功能。归脾丸溶液改善记忆功能可能与改善造血及海马神经元功能有关。

     

    Abstract: Objective To observe the effect of Gui-pi decoction on memory function in mice exposed to benzene and explore possible mechanism of Gui-pi decoction in memory function improvement in benzene exposed mice.

    Methods Seventy two Kunming mice were randomly selected into 6 groups:the blank control group, the benzene exposure only group, the routine treatment group(Leucosonurn and Batilol) and the Gui-pi decoction groups in three doses(2, 4 and 8 mg/kg& #183;d). Each group contained 12 mice. A Morris water maze systems was applied to study mice behavior. Neuronal morphology and blood tests were also performed.

    Results The time of platform navigation and searching in the morris water maze test were significantly lower in the Gui-qi decoction groups than the benzene exposure only group (P<0.01). The number of cross-platform and swimming time in the target zone were significantly lower in the large dose Gui-pi group than in the benzene ezposure only group. The nerve cell morphology and count of hippocampus were significantly different. The hemoglobin were higher in the large and medium dose Gui-pi groups than in the routine treatment group (P<0.05).

    Conclusion Gui-pi decoction may improve memory function in benzene exposed mice, which could contributed by improving hematopoiesis and hippocampal neurons function.
