锌-金属硫蛋白对PM2.5致运动大鼠氧化-免疫损伤的拮抗作用

Antagonism Effect of Zinc-Metallothionein to Antioxidation-Immune Damage Induced by PM2.5 in Exercising Rats

  • 摘要: 目的

    探讨锌-金属硫蛋白(Zn-MT)对PM2.5暴露致运动大鼠肺组织氧化损伤及部分免疫功能下降的拮抗作用。

    方法

    选取雄性SD大鼠32只,随机分为运动对照组、PM2.5染毒组、PM2.5+运动组、Zn-MT+PM2.5+运动组,在一次递增负荷跑台训练后即刻处死,取肺组织制成匀浆后测定氧化应激指标丙二醛(MDA)、过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)及谷胱甘肽过氧化物酶(GSH-Px)和免疫指标白细胞介素-2(IL-2)、IL-6、IL-8及单核细胞趋化蛋白1(MCP-1)。

    结果

    与运动对照组相比,PM2.5染毒组IL-6、IL-8、MCP-1均升高,GSH活性下降(P<0.05);与PM2.5染毒组相比,PM2.5+运动组大鼠肺组织GSH-Px下降(P<0.01),MDA、IL-2、IL-6上升;补充Zn-MT后,与PM2.5+运动组比较,CAT、GSH呈回升趋势,IL-6、IL-8有所下降。

    结论

    PM2.5暴露在一定程度上可以造成机体抗氧化稳态紊乱和免疫力的破坏,Zn-MT在一定程度上可拮抗由于PM2.5暴露导致的肺抗氧化损伤和免疫系统损伤。

     

    Abstract: Objective

    To examine the antagonism effect of zinc-metallothionein (Zn-MT) on the antioxidation-immune damage in exercising rats exposed to fine particulate matters (PM2.5).

    Methods

    Thirty-two male SD rats were randomly assigned into exercise control group,PM2.5 exposed group,PM2.5+exercise group,and Zn-MT+PM2.5+exercise group.All rats were immediately executed after one-time treadmill training with increasing loads and collected lung tissue homogenate samples for measuring oxidative stress indicators including malondialdehyde (MDA),catalase (CAT),glutathione (GSH) and glutathione peroxidase (GSH-Px),and immunity indicators including interleukin-2(IL-2),interleukin-6(IL-6),interleukin-8(IL-8),and monocyte chemoattractant protein-1(MCP-1).

    Results

    The levels of IL-6,IL-8,and MCP-1 increased but the activities of GSH declined in the PM2.5 exposed group,compared with the exercise control group (P<0.05).The activities of GSH-Px decreased (P<0.01) but the levels of MDA,IL-2,and IL-6 increased in the PM2.5+exercise group compared with the PM2.5 exposed group.After adding Zn-MT,compared to the PM2.5+exercise group,CAT and GSH showed a rising trend but IL-6 and IL-8 declined.

    Conclusion

    Zn-MT could antagonize the antioxidant and immune injury due to PM2.5 exposure in exercising rats.
