Urinary 8-OHdG Levels and Related Impact Factors in General Population

  • 摘要: 目的

    探讨一般人群尿8-羟基脱氧鸟苷(8-OHdG)水平,以及可能影响因素的作用。

    方法

    选择华东某市的城市居民共257名作为研究对象,采用超高效液相色谱-串联质谱方法测定尿中8-OHdG水平,高效液相色谱法检测尿中肌酐的浓度,校正尿中的8-OHdG的浓度。影响因素分析采用多重线性回归分析,交互作用采用多因素方差分析模型分析。

    结果

    该地区一般人群尿中的8-OHdG的质量浓度MP25P75)为1.50(0.39~2.24)μg/L,肌酐校正后MP25P75)为129.82(50.79~235.58)μg/mol。经吸烟、饮酒情况和体质指数(BMI)分层后,没有发现吸烟、饮酒和BMI单独对尿中8-OHdG改变的影响。多重线性回归分析结果显示,在排除其他混杂因素的影响后,饮酒对尿中8-OHdG水平有影响(P=0.040)。进一步分析发现超重人群中随年龄升高8-OHdG水平有升高趋势(Ptrend < 0.05)。

    结论

    年龄与BMI的交互作用是尿中8-OHdG水平的重要影响因素,尤其是高年龄的超重者更易发生DNA氧化损伤。

     

    Abstract: Objective

    To measure urinary 8-hydroxy-2'-deoxyguanosine (8-OHdG) levels in general population and assess the roles of potential impact factors.

    Methods

    The urinary levels of 8-OHdG of 257 urban residents in a city of east China were measured with ultra-high performance liquid chromatography-mass spectrometer, and creatinine by high performance liquid chromatography to adjust the urinary 8-OHdG levels. The impact factors were analyzed by multivariate linear regression models, and interaction effects by multivariate analysis of variance.

    Results

    The median (P25~P75) level of 8-OHdG in local general population was 1.50 (0.39-2.24) μg/L, and the level adjusted by creatinine in urine was 129.82 (50.79-235.58) μg/mol. We did not find changes of urinary 8-OHdG by smoking, drinking, and body mass index (BMI) alone after stratification analyses. By multivariate linear regression analysis, we found that drinking had a significant effect on the urinary levels of 8-OHdG (P=0.040) after controlling other selected confounding factors. And we found an increasing trend of urinary levels of 8-OHdG with older age in the overweight group (Ptrend < 0.05).

    Conclusion

    The interaction between age and BMI might be an important impact factor for urinary levels of 8-OHdG. Overweight individuals in older age group tend to be at a higher risk of DNA oxidative damage.
