2,4-二异氰酸甲苯酯暴露对小鼠血清中特异性IgG抗体水平的影响

Effects of 2,4-Toluene Diisocyanate Exposure on Levels of Serum Specific IgG Antibody in Mice

  • 摘要: 目的 探讨不同剂量和时间2,4-二异氰酸甲苯酯(2,4-TDI)气体暴露对小鼠血清中特异性IgG抗体水平的影响。

    方法 将清洁级(SPF) BALB/c雌性小鼠64只,按照染毒时间7 d和14 d随机分为两部分,共8组,每组8只,每部分分别设对照组及低(0.036 mg/m3)、中(0.14 mg/m3)、高(0.36 mg/m3)浓度暴露组,采用TDI连续动式吸入染毒,每天染毒4 h,记录体重,于末次染毒24 h采血分离血清,采用酶联免疫法检测特异性IgG含量。

    结果 暴露7 d时,3个暴露组小鼠血清特异性IgG抗体含量(光密度值)分别为(0.14& #177;0.0084),(0.14& #177;0.0064)及(0.14& #177;0.0076),与对照组(0.14& #177;0.0093)相比,差异无统计学意义(P > 0.05);连续暴露14 d后小鼠血清中特异性IgG抗体含量,各组间差异有统计学意义(F=11.41,P < 0.05),进一步两两比较分析,中、高暴露组血清中特异性IgG抗体含量分别为(0.17& #177;0.0200)和(0.18& #177;0.0210),均高于对照组(0.14& #177;0.0046)(P < 0.05),高暴露组血清中特异性IgG抗体含量高于低暴露组(0.15& #177;0.0120)(P < 0.05);中暴露组的暴露14 d小鼠血清中特异性IgG抗体含量为(0.17& #177;0.0200),高于暴露7 d (0.14& #177;0.0064)(P < 0.05);高暴露组的暴露14 d小鼠血清中特异性IgG抗体含量为(0.18& #177;0.0210),高于7 d (0.14& #177;0.0076)(P < 0.05)。

    结论 TDI气体连续暴露浓度及时间均影响小鼠血清中特异性的IgG抗体水平的表达。

     

    Abstract: Objective To examine the effects on serum specific IgG antibody level in mice exposed to 2,4-toluene diisocyanate (2,4-TDI) at different doses and for different time.

    Methods Specific pathogen free (SPF) BALB/c mice (female,n=64) were randomly divided by the exposure time of 7 or 14 days into two parts with a total of eight groups containing eight rats per group.Each part included control group,low (0.036 mg/m3),middle (0.14 mg/m3),and high (0.36 mg/m3) dose groups.Rats in the latter three groups were continuously exposed to 2,4-TDI vapor for 4 h/d.Weight was recorded and serum samples were collected 24 h following the final exposure.The levels of serum specific IgG were detected by enzyme-linked immunosorbent assay.

    Results The average optical density level of specific IgG antibody in each dose group on day 7(0.14& #177;0.008 4,0.14& #177;0.006 4,0.14& #177;0.007 6) had no significant difference compared with the control group (0.14& #177;0.009 3)(P < 0.05).And the levels of specific IgG antibody on day 14 were different among the groups (F=11.41,P < 0.05).The results of pair-wise comparison showed that the middle dose group (0.17& #177;0.020 0) and the high dose group (0.18& #177;0.021 0) presented higher levels of specific IgG antibody than the control group (0.14& #177;0.004 6)(P < 0.05),and the high dose group also presented higher level of specific IgG antibody than the low dose group (0.15& #177;0.012 0)(P < 0.05).The middle dose group on day 14(0.17& #177;0.020 0) showed a higher level of specific IgG antibody than on day 7(0.14& #177;0.006 4)(P < 0.05).Similarly,the high dose group showed higher level of specific IgG antibody on day 14(0.18& #177;0.021 0) than on day 7(0.14& #177;0.007 6)(P < 0.05).

    Conclusion Both concentration and time of TDI continuous exposure could influence the expression levels of serum specific IgG antibody.
