三代小鼠十溴联苯醚灌胃染毒对子鼠(F1、F2)学习记忆能力的影响

Effects of Decabromodiphenyl Ether Exposure by Gavage on Learning and Memory in F1 and F2 Offspring Mice

  • 摘要: 目的 考察三代小鼠十溴联苯醚(decabromodiphenyl ether,BDE 209)灌胃染毒对两代子鼠空间学习记忆能力和空间位置记忆能力的影响。

    方法 将75 只雌性昆明F0 代小鼠随机分为对照组0 mg/(kg& #183;d)、1.5 mg/(kg& #183;d)和225 mg/(kg& #183;d)染毒组,每组25 只,BDE 209 灌胃10 d 后,与雄鼠合笼,每组选取怀孕时间相近(相差不超过2 d)的8 只母鼠孕期持续染毒至子鼠断乳。第一代(F1)及第二代(F2)子鼠在断乳后也按照与母鼠相同的方式进行染毒。观察出生后子鼠数量并测量第30、60 和90 天子鼠体重,并利用Morris 水迷宫评价出生后第30、60 和90 天子鼠的空间学习记忆能力和空间位置记忆能力。

    结果 与对照组比较,1.5 mg/(kg& #183;d)组F0 和F1 代母鼠产仔数量分别降低约25%、30%(P<0.05);225 mg/(kg& #183;d)组F1 代出生后30 天雌性子鼠体重降低约17%(P<0.05);染毒组F1 代出生后30 天雄性子鼠体重分别降低约12%(P<0.05)、25%(P<0.01);染毒组F2 代出生30 天雌性子鼠体重降低约12%(P<0.01);1.5 mg/(kg& #183;d)组F2 代出生后30 天雄性子鼠体重降低约8%(P<0.05),225 mg/(kg& #183;d)组F2 代出生后60、90 天雄性子鼠体重均降低约13%(P<0.01);各组F1 代60、90 天,F2 代30、60 和90 天雌性子鼠逃避潜伏期延长,结果具有统计学意义(P<0.05 或P<0.01);F1 代1.5 mg/(kg& #183;d)组出生后60 天,各组出生后90 天,及F2 代225 mg/(kg& #183;d)组出生后30、60、90 天雄性子鼠逃避潜伏期延长,结果具有统计学意义(P<0.05 或P<0.01);F1、F2 代子鼠平台象限活动时间差异无统计学意义(P>0.05);F2 代出生后60 和90 天雌性子鼠平台穿越次数减少,结果具有统计学意义(P<0.05 或P<0.01);F2 代雄性子鼠出生后90天平台穿越次数减少,结果具有统计学意义(P<0.05)。

    结论 三代小鼠BDE 209 染毒能够影响子代发育,降低子代空间学习记忆能力和空间位置记忆能力。

     

    Abstract: Objective To evaluate the effects on learning and memory ability of two generations of offspring after prenatal and postnatal decabromodiphenyl ether (BDE 209) exposure by gavage.

    Methods Seventy-five Kunming female mice were randomly assigned to one control and two BDE 209 groups 1.5 mg/(kg& #183;d) and 225 mg/(kg& #183;d) through gavage for 10 days before mating. Then eight mice with close fertilization day were selected for each group and continued to gavage through pregnancy and lactation until weaning. Offspring in F1 and F2 were treated in the same protocol as the F0. Body weight and numbers of pups was recorded. Morris water maze was used to exam the ability of learning and memory in offspring at 30, 60 and 90 days after birth.

    Results The numbers of pups in F1 and F2 were significant decreased by 25% and 30% respectively in the low BDE 209 group (P<0.05), compared with the control group. The high BDE 209 exposure significantly decreased the body weight of the F1 30-day-old female pups by about 17% (P<0.05). The BDE 209 exposure significantly decreased the body weight of the F1 30-day-old male pups by 12% and 25%, respectively (P<0.05 or P<0.01). The BDE 209 exposure significantly decreased the body weight of the F2 30-day-old female pups by about 12% (P<0.01). The low BDE 209 exposure significantly decreased the body weight of the F2 30-day-old male pups by about 8% (P<0.05); the high BDE 209 exposure significantly decreased the body weight of the F2 60-and 90-day-old male pups by about 13% (P<0.01). The escape latency of the female F1 pups of 60 and 90 days old or the female F2 pups of 30, 60, and 90 days old was increased (P<0.05 or P<0.01). Also, the escape latency of the male F1 pups of 60 and 90 days old in the low exposure group or the male F2 pups of 30, 60, and 90 days old in the high exposure group was increased (P<0.05 or P<0.01). The time in target quadrant of the F2 female of 60 or 90 days old was decreased (P<0.05 or P<0.01). Also, the time in target quadrant of the F2 male of 90 days old was decreased (P<0.05).

    Conclusion Exposure to BDE 209 shows detrimental effects on developmental learning and memory in offspring mice.
