Study of Establishing the Cold Stress-Induced Osteoarthritis Model in Mouce

  • Objective To study the role of cold stress played in the etiology of osteoarthritis in murine for build a cold stress-induced osteoarthritis model in mouse.

    Methods Ten BALB/C mice a group in clean grade each were taken as model or control groups. The right knees of model group mice were stimulated with 6-8℃ water continuously for 30 days, 1.5 h/d. Then the degree of right knee swelling were measured with vernier caliper. The levels of TNF-α, rheumatoid factor(IgM) and hyaluronic acid (HA)in mice serum were detected with ELISA. The pathological slices of right knee were stained with hematoxylin-eosin(HE) dye for microscopy.

    Results When the experiment ended(day 31), the diameters of right knee of mice in model group were longer than that of left knee(P<0.01). Compared with that of initial day, a significant difference of the diameter of right knees was also observed. However, there were no such significant differences in control group(P>0.01). Significant increases of serum TNF-α and HA levels were observed in the model group compared with the control group(P<0.01), while the level of IgM showed no significant difference(P>0.05). A series of pathological changes were found in model group, including disordered articular cartilage cells showed:karyolysis or disappearing cellular nucleus, remarkable proliferation, calcification, adhesion of cartilage or synovial cells, and so on.

    Conclusion The cold stress could induce osteoarthritis in mice. The mice model of osteoarthritis was built preliminarily.
