Lipid Peroxidation Induced by Coal Tar Pitch Smoke Extract in Human Bronchial Epithelial Cells

  • Objective To explore whether coal tar pitch smoke extract (CTPE) could induce lipid peroxidation in human bronchial epithelial cells (BEAS-2B)

    Methods BEAS-2B cells were exposed to different concentrations (0,1,and 3 μg/mL) of CTPE for four hours and measured for the levels of reactive oxygen species (ROS) by flow cytometry.The amount of malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in the cells exposed to CTPE for eight hours were analyzed using MDA and SOD assay kits,respectively.

    Results The levels of ROS in the BEAS-2B cells treated with CTPE (1 and 3μg/mL) for four hours were higher than that in the control group.After eight hours exposure,the activity of SOD in the CTPE groups (1 and 3μg/mL) was significantly decreased compared with that in the control group;similarly,the amount of MDA and the ratio of MDA/SOD were also significantly higher than those in the control group.

    Conclusion CTPE can elevate levels of ROS and MDA,while decrease SOD activity.These results indicate that CTPE might induce lipid peroxidation in BEAS-2B cells.
