二甲基苯蒽卵巢被膜下注射诱发小鼠卵巢肿瘤模型

A Murine Ovarian Tumor Model Induced by 7,12-Dimethylbenzaanthracene Injection in Mice

  • 摘要: 目的 以二甲基苯蒽(7, 12-dimethylbenzaanthracene, DMBA)卵巢被膜下注射诱发小鼠卵巢肿瘤,并初步观察其生物学特性,为探讨卵巢癌的发病机制及评价治疗措施提供理想的动物模型。

    方法 取雌性C57BL/6小鼠10只,在麻醉状态下,用眼科微量注射器分别在小鼠双侧卵巢被膜内注入含有4 g/L的DMBA注射液0.05 mL,连续观察4个月,进行肿瘤生物学特性的一般状况观察、光镜观察、病理切片检查。

    结果 在注射DMBA后4个月发现, 1只小鼠因术后感染而死亡, 2只无肿瘤发生,其余小鼠出现腹部隆起,发现有不同程度的未分化癌或腺癌,组织学特点与相应的人类卵巢肿瘤很相似。

    结论 DMBA能原位诱发小鼠的卵巢肿瘤,肿瘤发生率达70%(7/10),可用于人卵巢癌的相关实验研究。

     

    Abstract: Objective To establish an ovarian tumor model in C57BL/6 mice with 7,12-dimethylbenzaanthracene (DMBA) injection and observe its biological features.

    Methods Ten female mice were injected with DMBA under ovary membrane through microinjector in operation and under aenethsia.Necropsy after 4months observation,and then gross aspects,light microscopy,and other respects of the cancer bearing mice were examined.Preliminary observations on the biological characteristics of the tumor tissue were performed.

    Results One mouse was died of severe infection after operation;two mice were tumor free,while the other seven were found bearing with undifferentiated carcinoma and/or aden carcinoma,respectively.Its biological features were quite comparable with ovarian tumor in woman.

    Conclusion DMBA can induce ovarian tumor in C57BL/6 mice and it can serve as an animal model for study of ovarian tumor.
