Hydroquinone Induced Changes in DNA Methylation and Potential Mechanism

    苯是一种广泛使用的工业原料,同时是一种确定的遗传毒性致癌物,主要由其活性代谢产物——氢醌发挥毒性作用。氢醌本身普遍应用于黑白显影剂、橡胶防老剂、稳定剂和抗氧剂。氢醌可引起机体多系统毒性损伤,长期试验表明氢醌还是一种致癌剂。近年来有文献报道氢醌暴露可导致DNA甲基化水平发生改变,本文综述了氢醌可能通过调节DNA甲基化转移酶的活力、氧化应激损伤干扰、DNA损伤修复蛋白调控以及其他表观遗传学修饰的参与等影响DNA甲基化状态,旨在为预防氢醌乃至苯中毒提供参考依据。

     

    Abstract:

    Benzene is widely used in industry and also a known genotoxic human carcinogens. Its toxicity is mainly caused by its active metabolite-hydroquinone. Hydroquinone itself is often applied in the black and white contrast medium, rubber protective agent, stabilizer, and antioxidant. Hydroquinone can cause multi-system damage to the body, even be a carcinogen in long-term tests. In recent years, hydroquinone exposure is reported to lead to changes of DNA methylation level. This paper reviewed that hydroquinone could affect DNA methylation by multiple mechanisms including regulation of DNA methylation transferase, oxidative stress injury interference, regulation of DNA damage repair proteins, and participation of other epigenetic modification, aiming to provide reference for prevention of hydroquinone and benzene poisoning.
