孕前慢性应激后仔鼠学习记忆与胰岛素样生长因子Ⅱ表达相关性研究

Association of Learning and Memory with Expression of Insulin-Like Growth Factor-Ⅱ in Offspring Rats Following Progestational Maternal Chronic Stress

  • 摘要: 目的 观察孕前慢性应激后仔鼠学习记忆能力与海马中胰岛素样生长因子Ⅱ(insulin-like growth factor-Ⅱ, IGF-Ⅱ)的表达变化,探讨仔鼠学习记忆能力变化的机制。

    方法 建立慢性应激孕前大鼠模型,通过血浆皮质酮动态测定来检测模型,采用Morris水迷宫和Y迷宫进行学习记忆能力测定、实时荧光定量聚合酶链式反应(real-timepolymerase chain reaction, RT-PCR)检测海马中IGF-Ⅱ表达水平。

    结果 模型组母鼠血浆皮质酮水平增高(P<0.05);模型组所产仔鼠只数及母鼠孕天数均少于对照组(P<0.05),两组雌雄比例差异无统计学意义(P> 0.05);模型组仔鼠血浆皮质酮高于对照组仔鼠(P<0.05);模型组仔鼠学习、记忆能力低于对照组(P<0.05);模型组仔鼠IGF-Ⅱ mRNA表达量低于对照组仔鼠(P<0.05)。

    结论 孕前慢性应激后仔鼠空间学习记忆能力下降,可能与其脑内IGF-Ⅱ表达的下调及体内皮质酮升高有关系。

     

    Abstract: Objective To observe the learning and memory abilities and the insulin-like growth factor-Ⅱ (IGF-Ⅱ) expressions in hippocampus of offspring rats after maternal rats exposed to stress before pregnancy, and to explore the mechanism of learning and memory abilities' alteration in offspring rats.

    Methods A maternal rat model was established by using chronic mild stress stimulation protocol. Plasma cortisol levels of the maternal model rats were measured by radioimmuneassay. The abilities of learning and memory were determined using Morris water maze and Y maze, and the expressions of IGF-Ⅱ by real-time polymerase chain reaction.

    Results Compared with the control maternal rats, the plasma cortisol levels of the maternal model rats were increased (P<0.05). Compared with the controls, the number of the model offspring rats and the days of maternal pregnancy in pregnant rats were less (P<0.05), but the male-to-female ratio was similar between the two groups (P> 0.05). The plasma cortisol levels of the model offspring rats were increased (P<0.05), while the learning and memory abilities (P<0.05) and the GF-Ⅱ mRNA expression levels (P<0.05) of the model offspring rats were decreased.

    Conclusion The progestational maternal stress exposure may reduce the learning and memory abilities in the offspring rats, which may be associated with the decreased expression of IGF-Ⅱ and the increased plasma cortisol level.
