Effect of tributyltin on body fat of zebrafsh

  • Objective To study on the effect of tributyltin (TBT) on body fat of zebrafish.

    Methods AB zebrafish larvae at 7 days post-fertilization (dpf) were divided into a blank control group, a solvent control group, and three TBT groups, with 100 larvae in each group. The three TBT groups were exposed to 12.5, 25.0, and 50.0 ng/L TBT until 30 dpf, respectively. Then body weight, length, body mass index (BMI), triglyceride (TG), and total cholesterol (TC) of each group were measured or calculated. HE staining and frozen oil red O staining were used to observe liver histological changes and hepatic steatosis of zebrafish samples.

    Results The body weight, BMI, and TG in the 25.0 ng/L and 50.0 ng/L TBT groups were significantly higher than those in the blank control group (P < 0.01). The body weights were (2.66±0.33), (3.72±0.58), and (1.16±0.25) mg, respectively; the BMI were (0.07±0.02), (0.10±0.03), and (0.03±0.01)mg/mm2, respectively; the TG levels were (24.41±0.80), (29.47±0.79), and (17.87±0.50) μmol/g (per g of protein), respectively. No difference in TC was found between the TBT groups and the blank control group. Hepatic steatosis was observed in the samples of the 25.0ng/L and 50.0ng/L TBT groups. Deeper liver oil red O staining in the 50.0ng/L group was observed compared with the blank control group.

    Conclusion TBT could increase the fat level of zebrafish, showing an obesogenic effect.
